The Hollywood Reporter reports that the new all-female Ghostbusters movie will star Kristen Wiig, Melissa McCarthy, Kate McKinnon, and Leslie Jones.
McCarthy and Wiig join the cast with one major blockbuster comedy together under their belt—Bridesmaids— but neither McKinnon nor Jones have starred in a movie of this caliber. They are, both, however, hilarious. Both are currently on Saturday Night Live, and McKinnon won an Emmy for her work on the show last year.

Paul Feig, who created Freaks & Geeks and directed Bridesmaids, announced on Twitter in October that he would be creating another Ghostbusters movie, but this time it will feature female leads:
It's official. I'm making a new Ghostbusters & writing it with @katiedippold & yes, it will star hilarious women. That's who I'm gonna call.

The rumor of this movie has been circulating since early August. Bill Murray — star of the original Ghostbusters movie — gave his top picks for an all-female cast: Kristen Wiig, Melissa McCarthy, Linda Cardellini, and Emma Stone.
It looks like Murray got 2 out of his 4 picks. A release date hasn't been set yet, but early indications suggest it'll be worth the wait in any case.
